CAMK2A / CaMKII Alpha Antibody - Anti-CaM Kinase II Antibody - Western Blot. Western blot of Affinity Purified anti-CaM Kinase II antibody shows detection of a band ~54 kD corresponding to human alpha CaM Kinase II (arrowhead lane 1). Specific reactivity with this band is blocked when the antibody is pre-incubated with the immunizing peptide (lane 2). Approximately 35 ug of a mouse brain whole cell lysate was separated by 4-20% SDS-PAGE and transferred onto nitrocellulose. CaM Kinase II was similarly detected on lysates from rat brain (not shown). After blocking the membrane was probed with the primary antibody diluted to 1:1500 for 2h at room temperature followed by washes and reaction with a 1:10000 dilution of IRDye800 conjugated Gt-a-Rabbit IgG [H&L] MX ( for 45 min at room temperature. IRDye800 fluorescence image was captured using the Odyssey Infrared Imaging System developed by LI-COR. IRDye is a trademark of LI-COR, Inc. Other detection systems will yield similar results.

Description
CAMK2A (calcium/calmodulin-dependent protein kinase II alpha) belongs to the Ca(2+)/calmodulin-dependent protein kinase family, and it is involved in plasticity at glutamatergic synapses, hippocampal long-term potentiation and spatial learning. CAMK2A mutations have been associated with autism spectrum disorder (ASD), impaired physical and intellectual development, and seizures. For example, expressed E183V-mutant CAMK2A protein has reduced targeting of dendritic spines, which causes a reduction in density and excitatory synaptic transmission. This is associated with the behavioral changes seen in ASD including repetitive behavior and impaired social interaction. In immunohistochemistry, CAMK2A is cytoplasmic and membranous positivity in neurons, the adrenal gland, the lung, the liver, the pancreas, the gastrointestinal tract, the kidney, reproductive tissues, and immune tissues. This affinity purified antibody was prepared from whole rabbit serum produced by repeated immunizations with a synthetic peptide corresponding to an N-terminus region of Human CaM Kinase II protein.

Specificity
A BLAST analysis was used to suggest reactivity with this protein from human, mouse, rat, and orangutan based on 100% homology for the immunogen sequence. Cross reactivity with CaM Kinase II protein from zebrafish and rabbit may occur as this sequence only varies by one amino acid residue (94% homology). Cross reactivity with CaM Kinase II homologues from other sources has not been determined.

Usage
Immunohistochemistry: LS-B334 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B334 was determined to be 2.5 ug/ml.
