Neutralization of bioactivity, Direct ELISA, Western Blot, Flow Cytometry. Neutralization of Mouse IL-15 R Bioactivity: The exact concentration of antibody required to neutralize the mouse cell surface IL-15 R mediated bioactivity is dependent on the cytokine concentration, cell type, growth conditions and the type of activity studied. The Neutralization Dose50 (ND50) for this antibody is defined as that concentration of antibody required to yield one-half maximal inhibition of the cell surface IL-15 R mediated IL-15 response on a responsive cell line, at a specific IL-15 concentration. As shown in Figures 1 and 2 on the next page, the ND50 for this lot of anti-mouse IL-15 R antibody was determined to be approximately 0.3-1.5 ug/mL in the presence of 0.8 ng/mL of human IL-15, using the CTLL-2 cell proliferation assay. Direct ELISA: This antibody can be used at 0.5-1.0 ug/mL with the appropriate secondary reagents to detect mouse IL-15 R. The detection limit for rmIL-15 R is approximately 0.1 ng/well. In this format, this antibody shows approximately 5% cross-reactivity with rhIL-15 R and less than 1% cross-reactivity with rhIL-2 R, rmIL-2 R and rmIL-2 R. Western Blot: This antibody can be used at 0.1-0.2 ug/mL with the appropriate secondary reagents to detect mouse IL-15 R. The detection limit for rmIL-15 R is approximately 5 ng/lane under non-reducing and reducing conditions. Flow Cytometry: This antibody can be used at 3-10 ug/mL/1E6 cells with an appropriate secondary antibody for indirect immunofluorescence staining of cells by flow cytometry.
