Recombinant soluble human CD66b.

Antibody from clone ANC3C5 binds to CD66b on the cell surface of human PMN in Flow cytometry and works as an EIA capture or detection reagent for recombinant CD66b when paired with clone ANC4A4.

Five x 105 Polymorphprep prepared human Granulocytes(PMN) per tube were pelleted and pre incubated 5 minutes with 20ul of human IgG at 300ug/ml (to reduce non specific binding) after which they were washed and incubated 45 minutes on ice with 80 ul of anti-CD66b antibody(ANC3C5) at 5 ug/ml. Cells were washed twice and incubated with 50 ul of 2o reagent Streptavidin/PE, after which they were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.66 log10 fluorescent units when compared to a Mouse IgG1/Biotin isotype control. Binding was blocked when cells were pre incubated with an excess of unlabeled anti-human CD66b (ANC3C5) mAb. The applications listed have been tested for the unconjugated form of this product. Other forms have not been tested.
