Farnesoid X receptor (FXR), a NR1 Thyroid Hormone-Like Receptor, has been shown to affect bile-acid metabolism in the liver and intestine. FXR binds to target DNA as a heterodimer (with retinoid X receptors) and is activated by bile acids. Binding of bile acids to the RXR/FXR heterodimer in liver represses CYP7A1 expression and bile-acid production, leading to a failure to solubilize and absorb cholesterol. In the intestine, binding of bile acids to the RXR/FXR complex activates the intestinal bile acid-binding protein (IBABP) and results in increased recycling of bile acids. A pseudogene of FXR, located on chromosome 1 (1p13.1 - 1p13.3), has been identified recently using a comparative genomics approach.
