Myotubularin-related protein 4 (MTMR4), a myotubularin-related phosphatase, has been shown to contain the consensus active site of dual specificity protein phosphatases (DSPs) and to share inhibition and inactivation characteristics with other DSPs. Similar to other myotubularin-related proteins, MTMR4 has been shown to possess a SET (Suvar3-9, Enhancer of zeste, trithorax)-interacting domain (SID) which may mediate interactions with SET domains present in chromatin-organizing nuclear proteins. Additionally, MTMR4 has been shown to possess a double zinc finger (FYVE) domain at the C-terminus and to efficiently and specifically dephosphorylate phosphotidylinositol 3-phosphate (PtdIns3P). Thus, MTMR4 is believed to be associated with the phosphatidylinositol 3-kinase pathway and membrane trafficking. Two mRNA splice variants have been observed.
