ERK3, a MAPK type protein kinase, has been associated with the ERK MAPKs based on similarity of the catalytic domain. Compared to ERK1/2, ERK 3 lacks the activating tyrosine phosphorylation site and contains only a serine which is phosphorylated by MEK. ERK3 is primarily localized to the nucleus where it is believed to phosphorylate substrates in response to cell stress and damage. Specific ERK3 substrates are uncertain; however, studies have shown that ERK3 does not phosphorylate ERK1/2 substrates. The ERK3 gene maps to chromosome 15q21, a region associated with chromosomal abnormalities in acute nonlymphoblastic leukemias. Additionally, ERK3 kinase activity has been reported to be increased in colorectal cancers.
