MAP4K4 is a MSN protein kinase. The protein has a structure similar to other GCK (Group I) kinases and contains two consensus SH3 binding motifs, which have been shown to interact with the SH3 domains of the SH2/SH3 adapter protein Nck. NIK can be activated by tumor necrosis factor (TNF)-alpha. Activated NIK has been shown to activate c-Jun N-terminal kinase (JNK) and to bind and activate the plasma membrane Na-H exchanger NHE1. In NIK(-/-) mice, mesodermal and endodermal cells of the primitive streak fail to migrate to their correct location, and embryos die between embryonic day (E) 9.5 and E10.5. At least three protein isoforms have been reported.
