GLK, a KHS protein kinase, activates the c-Jun N-terminal kinase (JNK) signaling pathway. Evidence suggests that it functions upstream of MEKK1. The protein has a structure similar to other GCK (Group I) kinases and contains an endophilin consensus-binding sequence PPRPPPPR, which has been shown to bind the SH3 domain of endophilin I in rat. GLK is activated by UV radiation and the proinflammatory cytokine tumor necrosis factor (TNF)-alpha.
