Type 1 Angiotensin Receptors (AT1A and AT1B) mediate the major cardiovascular effects of angiotensin II, controlling blood pressure and volume by regulating cell growth, vascular contraction, inflammatory responses, and salt and water retention. Stimulation of AT1 receptors by Ang II activates phospholipase C beta, resulting in increased intracellular calcium and inositol 1, 4, 5-triphosphate concentrations, and activatesthe mitogen-activated protein kinases, such as extracellular regulated kinases, by way of Src and Ras as well as the JAK/STAT pathways. AT1 knock-out mice have lower blood pressure than wild type mice and lose normal pressor response to exogenous Ang II. AT1B differs from AT1A in its mRNA tissue distribution and in the 5' untranslated region. AT1 blockers have become widely used in the treatment of hypertension and may offer benefits other than blood pressure reduction.
