Predicted nonglycosylated monomeric weight: 40 kd. TIGIT-muIg runs as a dimer in SDS-PAGE with a molecular weight of approximately 100 kD. Five x 10^5 cultured U-937 human tumor cells were washed and pre incubated 10 minutes with 20ul of 300ug/ml human IgG (to reduce non specific binding), after which they were incubated 45 minutes on ice with 80 µl of TIGIT-muIg/Biotin at a concentration of 5 µg/ml. Cells were washed twice and incubated with secondary reagent Streptavidin/R-Phycoerythrin, after which they were washed twice, fixed and analyzed by FACS. Cells stained positive with a mean shift of 0.46 log10 fluorescent units when compared to Recombinant muIgFc/Biotin negative control. Binding was blocked when reagent was pre incubated with a >10 fold excess of recombinant CD155-muIg.
