A soluble molecule consisting of the mature extracellular (213 aa) domain of human CD275 (GL50,ICOS L) fused to the murine IgG2a Fc (233 aa). (449 aa total). The molecule is dimeric with a predicted monomeric non glycosylated molecular weight of 50.3 kd. Five x 10^5 human HPB-MLT cells per tube were washed and incubated 45 minutes on ice with 80 µl of CD275trn-muIg/Biotin at a concentration of 5 µg/ml. Cells were washed twice and incubated with secondary reagent Streptavidin/R-PE, after which they were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.44 log10 fluorescent units when compared to a Mouse IgG2a/Biotin negative control (catalog#281-030) at a similar concentration. Binding was blocked when reagent was pre incubated with a 10-fold excess of recombinant CD278(ICOS)-muIg.
