Description
TYRP1 is a melanocyte-specific protein involved in melanin synthesis. It functions to stabilize tyrosinase protein and modulate its catalytic activity. It also plays a role in the maintenance of melanosome structure and influences melanocyte proliferation and cell death. Mutations in this gene are associated with oculocutaneous albinism type 3 (OCA3). Expression of this protein in metastatic melanoma correlates with Breslow thickness and poor prognosis, and it may be an effective target of inhibition therapy. In immunohistochemistry of normal tissue, it has membranous positivity on melanocytes. Usage
Immunohistochemistry: LS-B4011 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B4011 was determined to be 5 ug/ml. Western Blot: Suggested dilution at 1µg/ml in 5% skim milk / PBS buffer, and HRP conjugated anti-Rabbit IgG should be diluted in 1: 50,000 - 100,000 as second antibody. ELISA titer in peptide based assay: 1:312500.
