MTOR Antibody - Anti-mTOR Antibody - Western Blot. Western blot of Affinity Purified anti-mTOR antibody shows detection of a band ~245 kD corresponding to human mTor (arrowhead). Approximately 30 ug of HEK293 cell lysate was separated by 4-8% SDS-PAGE and transferred onto nitrocellulose. After blocking, the membrane was probed with the primary antibody diluted to 1:650 for 2h at RT. The membrane was washed and reacted with a 1:10000 dilution of IRDye800 conjugated Gt-a-Rabbit IgG [H&L] MX ( for 45 min at room temperature. IRDye800 fluorescence image was captured using the Odyssey Infrared Imaging System developed by LI-COR. IRDye is a trademark of LI-COR, Inc. Other detection systems will yield similar results.

Description
MTOR (mechanistic target of rapamycin, FRAP1) is a serine/threonine protein kinase that acts as a central regulator of cellular metabolism, growth and survival in response to hormones, growth factors, nutrients, energy and stress signals. MTOR directly or indirectly regulates the phosphorylation of at least 800 proteins. Overexpression of MTOR is related to tumorigenesis in breast, lung, bladder, brain, renal, thyroid, and prostate cancer as well as melanoma and leiomyosarcoma, and it is a target of therapeutic inhibitors. In immunohistochemistry of normal tissue, MTOR has membranous and cytoplasmic positivity in epithelial tissues throughout the body including intestinal crypts, the isthmic region of the stomach, pancreatic ductal cells, liver bile ducts, the kidney distal nephron and the bladder. This affinity purified antibody was prepared from whole rabbit serum produced by repeated immunizations with a synthetic peptide corresponding to an internal region near amino acids 2430-2460 of human mTOR.
Specificity
A BLAST analysis was used to suggest cross reactivity with mTOR protein from rat and mouse based on 100% homology with the immunizing sequence. Expect partial reactivity against mTOR homologues from zebrafish (94%) and dog (89%). Reactivity against homologues from other sources is not known.

Usage
Immunohistochemistry: LS-B650 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B650 was determined to be 2.5 ug/ml.
