Recombinant Human CD155.

Antibody ANC2B2 binds to cell surface CD155 on U-937 cells, and binds to recombinant CD155 in EIA.

Five x 105 cultured human U-937 cells per tube were washed and pre incubated ~5 minutes with 20 ul human IgG at 300 ug/ml (to reduce non specific binding ) after which they were incubated 45 minutes on ice with 80 ul of anti-human CD155/Biotin at a concentration of 10 ug/ml. Cells were washed twice and incubated with 50 ul of 2o reagent GAM/FITC, after which they were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 0.69 log10 fluorescent units when compared to a Mouse IgG1/Biotin Isotype control. This binding was blocked when reagent was pre incubated with 100 ug/ml recombinant CD155-muIg.
