Immunohistochemistry: This antibody stained formalin-fixed, paraffin-embedded sections of human breast invasive ductal carcinoma. The recommended concentration is 2.5 ug/ml-5 ug/ml with an overnight incubation at 4°C. An HRP-labeled polymer detection system was used with a DAB chromogen. Heat induced antigen retrieval with a pH 6 sodium citrate buffer is recommended. Optimal concentrations and conditions may vary. Tissue samples were provided by the Cooperative Human Tissue Network, which is funded by the National Cancer Institute. ELISA: In a sandwich ELISA (assuming 100 ul/well), a concentration of 4-8 ug/ml of this antibody will detect at least 1000 pg/ml of recombinant human IL-2 when used with biotinylated antigen affinity purified anti-human IL-2 (
LS-C104522) as the detection antibody at a concentration of approximately 1-2 ug/ml. Western Blot: To detect hIL-2 by Western Blot analysis this antibody can be used at a concentration of 1-2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hIL-2 is 2-4 ng/lane, under reducing or non-reducing conditions.
