Usage
Suitable for use in Western Blot and Immunohistochemistry. Western Blot: 1:100-1:250. Immunohistochemistry: 1:100-1:300 in PBS - Bn/Av-HRP Technique. Control Tissue: The antiserum demonstrates positive labeling of rat cortex and. hippocampus. Perfusion Fixation: Fixative-4% paraformaldehyde in 0.1 M Phosphate buffer, pH 7.4; 500 mL over approximately 20 minutes. Post Fixation: 4% paraformaldehyde in 0.1 M Phosphate buffer, pH 7.4; 500 mL over approximately 20 minutes. Post Fixation-1.5 hour at 4? C. in 4% paraformaldehyde in 0.1 M phosphate buffer, pH 7.4. Note: Paraformaldehyde is a necessary component in fixation of serotonin. If needed, low levels of glutaraldehyde (0.10.3%) may be used in conjunction with paraformaldehyde. Sections: 50um vibratome. Antibody dilution: 1:100-1:300 in PBS-Bn-SA/HRP Technique. Absorption control: Synthetic rat 5HT 7 receptor. Incubation on Tissue: 16-24 hours at 4?C.
