Basigin / Emmprin / CD147 Antibody - IF analysis of CD147 using anti-CD147 antibody CD147 was detected in paraffin-embedded section of human intestinal cancer tissues. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution ) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1µg/mL rabbit anti-CD147 Antibody overnight at 4°C. DyLight®488 Conjugated Goat Anti-Rabbit IgG was used as secondary antibody at 1:100 dilution and incubated for 30 minutes at 37°C. The section was counterstained with DAPI. Visualize using a fluorescence microscope and filter sets appropriate for the label used.
Basigin / Emmprin / CD147 Antibody - Western blot analysis of CD147/Emmprin using anti-CD147/Emmprin antibody. Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: Human Hela whole cell lysates, Lane 2: Human Placenta whole cell lysates, Lane 3: Human Jurkat whole cell lysates, Lane 4: Human U20S whole cell lysates, Lane 5: Human K562 whole cell lysates, Lane 6: Human MCF-7 whole cell lysates, Lane 7: Human COLO-320 whole cell lysates, Lane 8: Human HepG2 whole cell lysates, Lane 9: Human PANC-1 whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CD147/Emmprin antigen affinity purified polyclonal antibody at 0.5 µg/mL overnight at 4°C, then washed with TBS-0.1% Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for CD147/Emmprin at approximately 38KD and 55KD. The expected band size for CD147/Emmprin is at 42KD.

E.coli-derived human CD147/Emmprin recombinant protein (Position: E138-A323). Human CD147/Emmprin shares 51.1% and 51.9% amino acid (aa) sequence identity with mouse and rat CD147/Emmprin, respectively.
Specificity
Present only in vascular endothelium in non- neoplastic regions of the brain, whereas it is present in tumor cells but not in proliferating blood vessels in malignant gliomas.
